Pharmacological and Toxicological Appraisal of Promising Enzyme-tyrosinase Isolated from Soil Bacteria-an Update

Aims: The membrane associated tyrosinase is an enzymatically active monomeric glycoprotein which purified to only a low degree. It has gained importance in the present era due its antioxidant and immunomodulatory properties as well applications industry. Moreover role synthesis of melanin skin for protection from UV radiations also paved way towards better understanding this enzyme.
 Study Design: Biochemical molecular characterization producing soil bacterium had been followed by assessment antioxidant, cytotoxic anti-cancer activities.
 Place Duration Study: Whole work completed at Microbiology Molecular biology labs IMBB during 2018- 2019.
 Methodology: Tyrosinase species were identified biochemical their genomic DNA sequencing BLAST analysis while crude was characterized Bradford's methods, activity total protein activity, on SDS-PAGE. free radical scavenging evaluated via DPPH, ABTS FRAP assays cell proliferation inhibition cytotoxicity calculated antitumor MTT assays.
 Results: P. putida, B. cereus, mycoides, M. luteus, K. pneumonia found be SDS-PAGE showed that mass about 39 kDa. Protein contents, specific highest mycoides [0.008±0.06 mg/ml, 1500±0.06 U/ml 346820.8±0.03 U/mg respectively]. results biological activities vary bacteria bacteria. putida higher [66.4±0.01% DPPH assay, 32.04±0.06%in assay 320.6±0.06 assay], [67.8±0.01%] [39±1.0% viability], Cereus [64.7±0.06% power 53.41±0.03% 159.6±0.06% 46.46±0.01% 43±0.75% viability].
 Conclusion: study revealed isolated different bacterial strains depicted optimal percentage anti-oxidative, anti-proliferative cellular viability can used near future medical industrial purposes.
